Pantoprazole Tablets: Complete Guide

Pantoprazole is a third-generation proton pump inhibitor (PPI) widely used for treating acid-related gastrointestinal disorders. Known for its superior stability at neutral pH and fewer drug interactions compared to other PPIs, pantoprazole has become one of the most commonly prescribed PPIs worldwide.

Mechanism of Action

Like all PPIs, pantoprazole irreversibly inhibits the gastric H⁺/K⁺-ATPase proton pump. However, pantoprazole has unique selectivity for cysteine 822 on the proton pump, compared to omeprazole which binds to cysteines 813 and 892. This selectivity contributes to pantoprazole’s more predictable pharmacokinetics and fewer CYP450 interactions.

  • Peptic ulcers: 40mg once daily for 4-8 weeks
  • H. pylori eradication: Part of triple or quadruple therapy regimens
  • Stress ulcer prophylaxis: 40mg IV in ICU patients
  • NSAID gastroprotection: 20-40mg daily
