India vs China Steroid API Sourcing for US Buyers

TL;DR

US buyers should source steroid or hormone APIs from India or China only after the exact manufacturer, site, batch controls, US import records, and filing support are documented. Choose India or China when one supplier clears the same evidence test; use dual sourcing only when sites, critical-material makers, and shipping lanes are genuinely separate.

Choose India when the Indian supplier clears your exact evidence test and the Chinese supplier does not. Choose China when the Chinese supplier clears that same test first, and use dual sourcing only when the two routes do not share the same critical-material maker, manufacturing site, or freight lane.

Country of origin helps you map supply risk. It does not prove that a steroid or hormone API fits your US program. FDA requires foreign API manufacturers to register and list commercially distributed APIs for US import, including the manufacturer’s API listing at entry. FDA import rules

Use This Country Selection Table First

Send the same request to every shortlisted supplier. Our award rule is simple: the supplier with the complete, product-specific evidence package wins the program.

A lower unit price cannot replace a missing manufacturer name, incomplete batch package, or unusable filing path. Use the table before samples, commercial negotiation, or an audit date.

What Evidence Matters More Than Country of Origin?

Start with the exact molecule, dosage-form plan, and intended US use. A steroid API for a US generic filing needs a different evidence package from a bulk drug substance intended for a compounding pathway.

Request these seven items in your first RFQ:

  1. Legal manufacturer and site-role map. Ask which site performs synthesis, purification, testing, packaging, and batch release.

  2. FDA import record. Ask for the foreign establishment’s FDA registration details, the API drug listing, and the named US importer. FDA says foreign API manufacturers must register and list commercially distributed APIs. FDA requirements

  3. Exact API specification. Ask for the salt, ester, polymorph where relevant, particle-size specification, analytical methods, and a recent batch-specific certificate of analysis.

  4. GMP and inspection evidence. Match the certificate’s site address and scope against the site that makes your proposed batch.

  5. Filing support. For an NDA, ANDA, or IND program, ask whether a Type II Drug Master File covers the exact API and whether the holder will issue a Letter of Authorization.

  6. Critical-material map. Ask for the manufacturer and country of origin of each critical starting material and key intermediate.

  7. Change and continuity terms. Ask who must notify you before a site, route, specification, test method, KSM, packaging, or lane changes.

A Drug Master File, or DMF, contains confidential manufacturing information that an applicant can reference. It is not FDA approval. FDA reviews a DMF only when an IND, NDA, ANDA, export application, or another DMF references it with the holder’s authorization. FDA DMF guidance

India vs China: Steroid API Risks

Steroid sourcing turns on containment and cleaning evidence. FDA Q7 says dedicated production areas should be considered for certain steroids with high pharmacological activity or toxicity, unless validated inactivation or cleaning procedures are established and maintained. FDA Q7 guidance

Ask both Indian and Chinese suppliers for the same four records:

  • The production-area and equipment map for the proposed steroid.
  • The cleaning-validation summary for shared equipment.
  • The impurity, residual-solvent, and assay methods used for the proposed lot.
  • The change-control process for route, equipment, and raw-material changes.

An India-made steroid API may still rely on China-origin inputs. India’s government reported that China accounted for 73.71% of India’s FY 2024-25 imports in listed API, bulk-drug, and intermediate categories. That figure does not identify the source of any individual steroid or hormone API. It tells buyers why the KSM map matters. India import data

A direct Chinese route can also work for a US program. The Chinese supplier must still identify the site, document the batch controls, meet US import requirements, and support the required filing path.

India vs China: Hormone API Risks

“Hormone API” is a buying category, not one manufacturing process. Your RFQ should identify the exact molecule before you compare countries.

For steroidal hormones, apply the steroid containment and cleaning review. For other hormone APIs, focus on the controls that define that molecule’s quality: identity, related substances, potency or assay, residual solvents where relevant, stability, and the validated method used for release.

Do not accept a category-level statement such as “hormone DMF available.” Ask whether the DMF, specification, batch method, and Letter of Authorization process apply to your exact API and proposed manufacturing site.

The same rule applies to pharmacopeial labels. USP, BP, or IP alignment can help define a material standard. Those labels do not identify the manufacturer, establish US import readiness, or show that a supplier’s current process matches your filing.

What US Importers Should Request First

Give your quality, regulatory, supply-chain, and importing teams one shared package. It should contain the following documents before the first commercial shipment:

  1. The manufacturer’s legal name, address, and site-role map.
  2. FDA foreign-establishment registration and the manufacturer’s API drug listing.
  3. The intended-use assessment and API labeling package.
  4. The batch specification, certificate of analysis, and analytical methods.
  5. The DMF number and LOA process when your US application requires filing support.
  6. The importer-of-record and customs-document responsibility matrix.
  7. The packaging configuration, storage conditions, and route-specific shipping evidence.
  8. The quality agreement and prior-notification terms for material changes.

FDA explains that imported APIs must meet applicable FD&C Act and 21 CFR requirements. FDA also states that APIs used in compounding must meet the applicable section 503A or 503B requirements. US API import guidance

Use FDA’s Import Alert search as part of the review. An alert search adds an independent check. It does not replace reviewing the supplier’s current site, batch, and change-control records.

When Dual Sourcing Is the Safer Strategy

Dual sourcing is safer when your two approved routes fail independently. That means separate manufacturing sites, separate critical-material manufacturers, separate release testing, and separate freight lanes.

Two suppliers in different countries are not independent when both buy the same KSM from one producer. They are also not independent when a single port, packer, test lab, or quality-release function can halt both supplies.

Build dual sourcing in this order:

  1. Qualify the primary supplier’s site and batch package.
  2. Define the second source’s matching specification and test methods.
  3. Map every critical material back to its manufacturer.
  4. Compare the two routes for shared sites, KSM makers, ports, and carriers.
  5. Approve the alternate source before a shortage or quality event forces the change.

FAQs

Is an FDA-Registered API Manufacturer FDA Approved?

No. FDA registration and drug listing are import and regulatory requirements. They do not mean FDA has approved the API for a particular finished drug product. FDA reviews the drug product application and any referenced DMF information in that application context. FDA DMF rules

Can a Certificate of Analysis Replace a Manufacturing-Site Review?

No. A COA reports results for one batch against a specification. A site review examines the controls that produce those results, including equipment, cleaning, records, deviations, and change management. FDA Q7 addresses those manufacturing controls across the API lifecycle. FDA Q7 guidance

How Much KSM Information Should a Buyer Request?

Request the name and address of the critical-material manufacturer, not only the supplier or trader. FDA Q7 says the API manufacturer should know the manufacturer’s name and address when a critical-material supplier is not that manufacturer. Use that information to find shared upstream dependencies. FDA Q7 requirements

Does a Type II DMF Cover Every Product a Supplier Sells?

No. A Type II DMF generally covers a single drug substance, intermediate, drug product, or material used in preparation. Ask whether the DMF covers your exact API, proposed site, and current process. FDA DMF types

Can the Same Qualification Package Support a Compounding Purchase?

No. The intended use changes the regulatory review. FDA states that APIs used in compounding must comply with the applicable section 503A or 503B requirements, so identify the compounding pathway before purchase. FDA compounding rules
