Bioconversion vs Chemical Synthesis Steroid API: Which Fits?
TL;DR
Bioconversion fits a steroid API when a proven biological step delivers the required selectivity with controlled conversion, by-products, recovery, and scale performance. Chemical synthesis fits when its material route, impurity purge, isolation, and technology transfer are better documented. Hybrid routes often win by using biology only where it solves a genuine chemistry problem.
Use five gates: the critical transformation, impurity fate, starting-material route, scale evidence, and transfer risk. Choose bioconversion when a proven enzyme or organism solves the critical transformation with manageable impurities.
Choose chemical synthesis when it has the stronger proven control package. Choose a hybrid route when biology solves one step and chemistry completes the API.
Choose the Route with Five Gates
Commercial steroid manufacture often combines microbial and chemical stages, so “bioconversion versus chemistry” is usually a decision about the critical step, not a facility category. A steroid biotechnology review describes microbial production of steroid intermediates followed by chemical modification into final APIs.
Use this table before asking a supplier for price or reserving a pilot campaign.
| Buyer Criterion | Bioconversion | Chemical Synthesis | Hybrid Route |
|---|---|---|---|
| Critical transformation | Selective hydroxylation or dehydrogenation | Established reaction sequence | Biological key step, chemical finish |
| Selectivity evidence | Exact substrate conversion data | Documented isomer control | Defined intermediate specification |
| Impurity review | Metabolite and conversion profile | Reaction and solvent profile | Handoff carryover profile |
| Material map | Sterol, substrate, catalyst sources | KSM, reagent, catalyst sources | Both maps, controlled intermediate |
| Scale evidence | Solubility, conversion, recovery | Heat, mixing, isolation | Hold time, recovery, transfer |
| Transfer controls | Strain, feed, isolation controls | Materials, CPPs, purification controls | Controls at every handoff |
| Supply continuity | Sterol and catalyst dependencies | Critical-material dependencies | Dependencies across both stages |
The table gives a practical answer. Select the route with the strongest evidence in the rows that create your product’s main risk.
Where Bioconversion Can Improve Selectivity and Impurity Control
Bioconversion uses whole cells or enzymes to convert a steroid substrate into a defined product or intermediate. It is most useful when one transformation needs difficult positional or three-dimensional selectivity.
A whole-cell CYP154C5 study produced 16α-hydroxylated products from six tested steroids. The same study detected small secondary-product amounts for androstenedione and nandrolone.
That distinction matters. A supplier cannot establish impurity control by calling a step enzymatic. The supplier must show what the exact substrate becomes, what remains unconverted, and what isolation removes.
Ask for these five records:
- A route summary that names the input steroid, biological transformation, and isolated intermediate.
- Representative conversion data from the intended substrate.
- Chromatograms that identify starting material, target, known by-products, and unknown peaks.
- The intermediate specification, including assay, related substances, water, and residual solvents where relevant.
- Batch data showing the same impurity pattern at development and intended production scale.
Microbial pathways can produce unexpected related compounds. A Rhodococcus study found C20-reduction products during cortisone and hydrocortisone biotransformations. That is why a buyer should assess the full conversion profile rather than a final purity percentage alone.
When the supplier sends a release package, use our steroid API COA checklist to compare the batch certificate with the approved specification and physical shipment.
When Chemical Synthesis Is the Better Manufacturing Route
Chemical synthesis is the stronger choice when the intended route has a proven impurity purge, qualified materials, reproducible isolation, and a defined transfer path to the commercial site.
Start with the impurity story. ICH Q11 expects manufacturers to understand impurity formation, fate, and purge through the process. A chemical route fits when the supplier can identify each critical impurity source and show where crystallisation, extraction, or another downstream operation controls it.
Choose chemical synthesis when the supplier can provide all four items below:
- A critical starting-material and reagent map.
- A process-flow summary with critical reaction and isolation steps.
- Representative impurity, residual-solvent, and yield trends.
- Commercial-scale equipment, batch-range, and transfer evidence.
A short route is not automatically a lower-risk route. A longer route can be the better commercial choice when its inputs, operating ranges, purification, and analytical methods are already controlled at the proposed manufacturing site.
How Hybrid Routes Are Used for Steroid APIs
Hybrid routes use biology where selectivity creates value, then use chemistry where chemistry offers the most controllable completion or derivatisation step. This approach matches how many steroid intermediates and APIs are made in practice. Published steroid research describes microbial sterol conversion followed by chemical synthesis of final steroid APIs.
The handoff between stages needs its own control strategy. Define the biological intermediate before it enters the chemical step.
Put these six fields in the supplier request:
- Intermediate name and chemical form.
- Release specification and analytical method.
- Related-substances profile.
- Storage condition and maximum hold time.
- Manufacturing and release site.
- Change notification for the biological or chemical stage.
A hybrid route wins only when the handoff is controlled. An excellent fermentation step does not help if the recovered intermediate changes during storage, purification, shipment, or transfer into the chemical sequence.
What Route Choice Means for Scale-Up and Supply Continuity
Bioconversion scale-up starts with substrate access to the biological catalyst. Steroids are poorly soluble in aqueous systems, which can limit conversion and productivity. A high-concentration dehydrogenation study achieved 92.5% hydrocortisone conversion within 6 hours at 80 g/L, illustrating why substrate concentration and catalyst performance must be shown with data.
Chemical scale-up starts with reaction and isolation behavior in the intended equipment. Review mixing, heat removal, addition sequence, reaction endpoint, crystallisation, filtration, washing, and drying.
For either route, request evidence from the exact equipment train. A 25 kg order quantity does not show validated batch range, reactor working volume, campaign frequency, or impurity reproducibility.
Supply resilience also follows the route. Map every critical dependency:
- Original manufacturer and country for each critical material.
- Qualified primary and alternate sources.
- Intermediate manufacturing and release sites.
- Planned inventory, lead time, and campaign capacity.
- Notice process for source, route, site, equipment, or method changes.
ICH Q7 requires a formal change-control system for changes that can affect intermediate or API production and control. It also treats a change in the source of critical raw materials under change control.
Before a pilot or commercial award, read our steroid API scale-up guide. It separates pilot feasibility from evidence that the intended commercial process can reproduce the API.
What US Buyers Should Request Before Selecting a Route
US buyers should select the route and supplier together. The route must match the exact API form, manufacturing site, quality system, and regulatory path.
Send this request to each shortlisted supplier:
For [API name and form], provide the proposed manufacturing site; a non-confidential route summary; critical-material and intermediate map; representative impurity and isolation data; intended batch range; current release specification; batch trend summary; change history; transfer plan; and Type II DMF scope and Letter of Authorization process where our filing will reference a DMF.
A Type II DMF can cover a drug substance, intermediate, or material used in preparing it. FDA explains that a Type II DMF supports confidential information for a referenced application.
A DMF is not a standalone approval. FDA reviews its technical content when an NDA, ANDA, IND, or other authorized submission references the file, as explained in the FDA DMF overview.
Use our steroid API RFQ checklist to collect matched answers on the molecule, site, release data, documentation, delivery, and change-control terms.
FAQs
Can a Supplier Keep the Route Confidential and Still Support Qualification?
Yes. A supplier can protect proprietary route details while providing a non-confidential process summary, site identity, specifications, impurity controls, and change-control commitments. For a US filing, the DMF holder can authorize FDA to review confidential information through a Letter of Authorization.
Does a High-Purity COA Prove the Route Is Controlled?
No. A COA shows batch-specific results against a stated specification. ICH Q7 requires API certificates to identify tests, acceptance criteria, results, and authorized approval, but route selection also needs process, impurity, scale, and transfer evidence.
Should a Change in Sterol Source Trigger a Quality Review?
Yes. Treat a changed critical sterol or starting-material source as a formal change-control event. Review the new material specification, impurity impact, comparability data, and any effect on the finished API control strategy.
When Should a Buyer Start a Second-Source Program?
Start when the API affects launch timing, patient supply, or a single-source critical intermediate. Our rule of thumb is to begin before supply pressure forces a route change, so the second source can generate comparable specification, impurity, stability, and documentation data.
